Design: After the sample DNA test was qualified, a certain proportion of lambda DNA was added as Bisulfite conversion test, and Covaris M220 was used to randomly interrupt genomic DNA to 200-400bp. End repair of broken DNA fragments, adding A tail, and methylated sequencing splices; Then Bisulfite treatment (using EZ DNA Methylation Gold Kit, Zymo Research) was performed. After treatment, the unmethylated C became U (T after PCR amplification), while the methylated C remained unchanged, and finally PCR amplification was performed. Get the final WGBS library

Study: Genome-wide DNA methylation of hindlimb muscles of Chinese alligator

We employed genome-wide bisulfite sequencing (WGBS) to generate single-base resolution DNA methylation maps of the hindlimb muscles in hibernating and active Chinese alligators, aiming to elucidate the regulatory mechanisms of DNA methylation in hibernation adaptation of Chinese alligators. The WGBS technique is widely regarded as one of the most effective methods for detecting DNA methylation. The findings from this study will provide a fundamental foundation for gaining further insights into the adaptive regulatory mechanisms of hindlimb muscles in Chinese alligators during hibernation.
